It’s now time to apply the skills and knowledge that you’ve developed to design, carry out, analyze and troubleshoot your own transfection/gene expression experiment!

Think about the results from your first experiment.  Did everything work as expected, without any technical issues?  Did you see an interesting result that is worth pursuing, either through a direct repeat or a variation on the first experiment?  Do you want to see if your results are reproducible in a different cell line?  Do you want to see if the drugs have the same effect on a different reporter?

Refer to the Transfection into Human Cells and the Luciferase Assay Protocols.  We will not have time to analyze protein expression through western blot analysis, but we can look for protein expression through fluorescence microscopy.
